Comparison of different tandem mass detection scans in the analysis of cannabidiol

The aim of this bachelor thesis was to compare different tandem mass spectrometer scans in order to develop an LC-MS/MS method suitable for the determination of cannabidiol in rat serum. In this bachelor thesis the mass spectrometer and high-performance liquid chromatography method were optimized. The optimum parameters were as follows: chromatographic column Acquity UPLC BEH C18 50 × 2,1 mm, 1,7 μm from Waters (Wexford, Ireland). The mobile phase consisted of methanol and water of LC-MS purity. The flow rate of the mobile phase was 0,4 ml/min, the column temperature 40 řC, the temperature in an autosampler 15 řC, the sample injection volume was 1 μl and the analysis time was 7 minutes under gradient elution conditions. The MRM transitions of highest intensity were monitored. For CBD, the MRM transition in positive mode was 315,2 → 193,1 (Q1 = -10 V, CE = -24 V, Q3 = -20 V), for isotopically labeled cannabidiol D3 was chosen 318,4 → 196,0 (Q1 = -10 V, CE = -23 V, Q3 = -20 V). The calibrations in solvent and rat serum were performed in MRM and SIM mode. No calibration was constructed for the SCAN mode due to its low selectivity, which made it able to detect cannabidiol only from concentration of 312,5 ng/ml. Physiological and pathological factors affecting absorption of drugs

Absorption of drugs from the gastrointestinal tract after oral administration is a key pharmacokinetic process co-determining the subsequent pharmacodynamic response of the organism and therapeutic efficacy. This dissertation thesis is devoted to the study of factors that influence this parameter. Special emphasis is placed on the study of lymphatic absorption, i.e. the rate of absorption of the active substance via the intestinal lymphatic system. A number of in vivo studies have been carried out in laboratory rats. Pharmacokinetic studies have been performed by means of regular blood sampling from vascular catheters after oral administration of the drug. Lymphatic absorption was investigated in an anaesthetized mesenteric lymphatic duct cannulated rat model. Modern drugs were tested that were incorporated into innovative dosage forms by collaborating chemical and technological institutions. Abiraterone acetate, a lipophilic agent used in the therapy of prostate cancer, was well absorbed from the gastrointestinal tract after administration in the form of oil marbles. Disposition and metabolism of cannabinoids.

This thesis describes in the form of a commentary on own original publications research on the problems of cannabinoids, ie. phytocannabinoids and some synthetic cannabinoids, their pharmacokinetics and effects. The work consists of four thematic areas: the pharmacokinetics of delta-9- tetrahydrocannabinol (THC) and cannabidiol (CBD) in rats, depending on the route of administration; THC concentration time profile in humans (after inhalation) and implications for transport safety; the pharmacokinetic profile of synthetic cannabinoids in rats; extraction and determination of phytocannabinoids in plant material. The first part of the thesis was to determine pharmacokinetic profiles of THC, CBD and combination thereof (1:1 weight ratio) in rats with respect to administration common in humans, i.e. inhalation, oral and subcutaneous administration. THC, its metabolites (11-hydroxy-tetrahydrocannabinol, 11-OH-THC; 11-nor-delta-9- carboxytetrahydrocannabinol, THCOOH) and CBD concentrations in serum and brains of animals were monitored at the 24 hours experimental interval during the study. Cannabidiol level in hemp (\kur{Cannabis sativa})

Hemp (Cannabis sativa) is a crop that people have grown for a lot of years ago. It is amazing by its almost versatile use, whether in medicine, food, industry and power engineering. The aim of this thesis was to evaluate the content of cannabidiol in different parts of hemp and the possibility of using these parts as a source of the compound. Morphologically distinct varieties - Bialobrzeskie and Finola were selected for the experiment. The Cannabidiol content was determined by high performance liquid chromatography (HPLC). Hemp roots, stems, male and female inflorescences and leaves were sampled for the determination of cannabidiol level. The highest content of cannabidiol was found in hemp flowers and leaves of Cannabis sativa. Therefore, theoretically, these parts of hemp grown for fiber could be used as a potential source of cannabidiol.
